CPNE6 Introduction
CPNE6 encodes copine-6, also known as neuronal copine or N-copine, a calcium-dependent phospholipid-binding protein distinguished by its predominantly brain-specific expression pattern. Within the central nervous system, CPNE6 transcripts and protein are most abundantly expressed in the hippocampus, with peak levels occurring during postnatal development — a temporal profile that coincides with the period of active synaptogenesis and dendritic spine maturation. Structurally, CPNE6 possesses the canonical copine architecture of two C2 domains and a von Willebrand factor A (vWA) domain, but it is unique among copine family members in its specialized neuronal functions. The C2 domains mediate calcium-triggered translocation from the dendritic cytosol to postsynaptic spine membranes, while the vWA domain engages downstream effector proteins involved in cytoskeletal remodeling and receptor trafficking.

The functional significance of CPNE6 was elucidated through landmark studies demonstrating its essential role in synaptic structural plasticity and hippocampus-dependent learning. Upon chemical induction of long-term potentiation, intracellular calcium transients recruit CPNE6 from the cytosol to the membranes of dendritic spines, where it translates the initial calcium signal into sustained structural changes including spine enlargement — a morphological correlate of synaptic strengthening. Mice lacking CPNE6 exhibit pronounced deficits in hippocampal long-term potentiation, impaired spatial and contextual learning, and reduced spine structural plasticity, establishing CPNE6 as a critical calcium sensor that bridges activity-triggered calcium signals to the morphological remodeling necessary for memory encoding. More recently, CPNE6 has been found to form a complex with AMPA-type glutamate receptors and to function as an escort protein for TRPM3 channels, influencing sensory neuron sensitivity. In melanocytes, CPNE6 contributes to dendrite formation, further highlighting its broader role in cellular morphogenesis beyond the nervous system. These findings collectively position CPNE6 as a key molecular integrator of calcium signaling, membrane dynamics, and structural plasticity in neurons.
