CPNE5 Introduction
CPNE5 encodes copine-5, a member of the copine family that shares the conserved structural organization of two N-terminal C2 domains and a C-terminal von Willebrand factor A (vWA) domain. Like other copines, CPNE5 is predicted to function as a calcium-dependent phospholipid-binding protein that translocates from the cytosol to membrane compartments in response to intracellular calcium fluxes, where it participates in calcium-mediated intracellular signaling and membrane trafficking events. The vWA domain of CPNE5 is thought to serve as a scaffold for assembling multiprotein complexes, though its specific binding partners and downstream effectors remain less well characterized compared to other copine family members. Notably, CPNE5 has been implicated in dendrite formation, suggesting a role in neuronal morphogenesis that parallels the brain-restricted expression pattern observed for several copine family paralogs.

In contrast to the oncogenic properties attributed to CPNE1, CPNE5 appears to function as a tumor suppressor in at least one clinically important context. In esophageal squamous cell carcinoma (ESCC), CPNE5 mRNA and protein expression are significantly reduced in tumor tissues compared with adjacent normal mucosa, and low CPNE5 expression independently predicts shorter overall survival following curative surgical resection. Patients whose tumors exhibit diminished CPNE5 levels experience higher rates of disease recurrence, particularly hematogenous metastasis, suggesting that CPNE5 loss may facilitate the metastatic cascade. The molecular mechanism by which CPNE5 suppresses tumor progression remains to be fully elucidated, but it may involve regulation of cell adhesion, cytoskeletal dynamics, or modulation of pro-metastatic signaling pathways through its C2 domain-mediated membrane interactions. Multivariable survival analyses have confirmed CPNE5 as an independent prognostic biomarker in ESCC, supporting its potential utility in postoperative risk stratification and clinical decision-making. The divergent roles of CPNE1 and CPNE5 in cancer — oncogenic versus tumor-suppressive — underscore the functional heterogeneity within the copine family and caution against generalized assumptions about family-wide biological behavior.
