BRMS1 Introduction

BRMS1 (breast cancer metastasis suppressor 1) is a member of a small family of proteins that suppress metastatic dissemination without affecting primary tumor growth. The protein is predominantly nuclear and acts as a scaffold that connects chromatin-modifying complexes to the SIN3–HDAC transcriptional repression machinery. The BRMS1 gene maps to chromosome 11q13.2 and is expressed in many tissues, with its loss frequently observed in metastatic breast cancer and melanoma.

Transcriptional Repression Through HDAC Recruitment
BRMS1 forms oligomeric complexes that recruit SIN3A–HDAC to specific promoters, leading to histone deacetylation and transcriptional repression of genes that drive metastasis, including osteopontin, uPA, and several chemokine receptors. Through these actions, BRMS1 reduces tumor-cell invasion, impairs intravasation, and restores gap-junction communication between tumor cells and stromal cells. BRMS1 also interacts with the nuclear factor E2F1 and represses NF-κB-dependent transcription, linking BRMS1 activity to cell-cycle control and to inflammatory signaling in the tumor microenvironment.

Metastasis Prognosis and Therapeutic Restoration
Low BRMS1 expression in primary breast and melanoma tumors correlates with increased risk of metastasis and shorter overall survival, supporting BRMS1 as a prognostic biomarker. Restoration of BRMS1 expression in metastatic cell lines reduces their ability to colonize distant organs, motivating efforts to identify small molecules that upregulate BRMS1 or mimic its activity. BRMS1 expression has also been shown to sensitize tumors to histone deacetylase inhibitors and to immune checkpoint blockade, opening potential combination therapy strategies for BRMS1-low cancers.
