BRIX1 Full Name
BRX1, biogenesis of ribosomes, homolog (S. cerevisiae)
BRIX1 Introduction
BRIX1 (biogenesis of ribosomes BRX1), also known as BXDC2 (Brix domain-containing protein 2), is a nucleolar protein that plays an essential role in the assembly of the large 60S ribosomal subunit. The gene encoding BRIX1 is located on human chromosome 5p13.2 and produces a protein of approximately 339 amino acids. First identified in Xenopus laevis as a highly charged protein with two putative nuclear localization signals, BRIX1 is evolutionarily conserved across eukaryotes, with orthologs found in yeast, C. elegans, Arabidopsis, Drosophila, and mammals. The name "BRIX" derives from "biogenesis of ribosomes in Xenopus," and its yeast homolog (BRX1, encoded by YOL077C) is an essential gene whose depletion leads to defects in rRNA processing and a block in large ribosomal subunit assembly. In humans, BRIX1 is ubiquitously expressed across tissues, with particularly high levels in the placenta and bone marrow.

Cellular Localization and Expression Pattern
BRIX1 is primarily localized to the nucleolus, the specialized subnuclear compartment where ribosomal RNA transcription, processing, and ribosome assembly take place. In transfected cells, BRIX1 also localizes to Cajal bodies (coiled bodies), nuclear organelles involved in the processing of small nuclear and small nucleolar RNAs. The protein is ubiquitously expressed across virtually all human tissues, consistent with its role as a general cellular "housekeeping" factor required for ribosome production in all cell types. The Human Protein Atlas confirms that BRIX1 is expressed in all tissues, with mainly nucleolar staining observed in most cell types. This ubiquitous expression pattern underscores the universal requirement for efficient ribosome assembly across diverse cell types and physiological contexts.
Molecular Function in Ribosome Biogenesis
The primary function of BRIX1 is to serve as an essential assembly factor for the 60S large ribosomal subunit. Through its RNA-binding activity, BRIX1 associates with 5S, 5.8S, and 28S ribosomal RNAs (rRNAs), facilitating the proper folding and maturation of these RNA components. In yeast, Brx1p is a constituent of 66S pre-ribosomal particles and is required for the processing of 25S rRNA and the assembly of large ribosomal subunits. The protein interacts with multiple other ribosome biogenesis factors, forming part of a complex that includes BRX1, RPF1, RPF2, and SSF1 or SSF2. The Brx1-Ebp2 complex plays a critical role in the early stages of large subunit assembly, as revealed by cryo-electron microscopy studies of early nucleolar pre-60S ribosomes at 3.7 Å resolution. This complex interferes with the assembly of specific rRNA domains, helping to orchestrate the stepwise construction of the ribosome's global architecture.
