BMP2K Full Name
BMP2 inducible kinase
BMP2K Introduction
BMP2K (BMP2-inducible kinase) is a serine/threonine kinase of the AMPK-related family that was originally identified as a transcript induced by bone morphogenetic protein 2 in osteoblast precursors. The protein contains a kinase domain, an autoinhibitory ubiquitin-associated (UBA) domain, and a C-terminal KA1 domain that mediates membrane association. BMP2K is broadly expressed and has been implicated in clathrin-mediated endocytosis and in transcriptional regulation in hematopoietic cells.

Endocytosis and Clathrin-Coated Pit Dynamics
BMP2K phosphorylates components of the endocytic machinery, including the clathrin-coat accessory proteins AP2 and ITSN1, supporting the scission of clathrin-coated pits and the internalization of receptor tyrosine kinases. The kinase activity of BMP2K is regulated by its UBA and KA1 domains, which respond to membrane curvature and to ubiquitin-dependent signals. In addition, BMP2K translocates to the nucleus in response to certain stimuli, where it phosphorylates transcription factors and modulates gene expression programs linked to cellular adaptation.
Leukemia and Receptor-Tyrosine-Kinase-Driven Cancers
BMP2K is highly expressed in acute myeloid leukemia and in chronic myeloid leukemia, where its activity sustains leukemic cell proliferation and may cooperate with BCR–ABL signaling. BMP2K has also been shown to modulate the surface expression of receptor tyrosine kinases such as EGFR and PDGFR, suggesting that BMP2K inhibition could affect the response to targeted therapies that depend on receptor internalization. Selective BMP2K inhibitors are in early preclinical development, with a focus on hematologic malignancies.
