Purification and Characterization of Exosomes

ISOLATION OF EXOSOMES

1. Preparing Exosome Collection Media

2. Pre-Centrifugation

3. Ultracentrifugation (or Alternative Methods)

CHARACTERIZATION OF EXOSOMES

1. Regular EV characterization

Total Protein Content: Perform BCA assay.
Particle Analysis: Measure particle concentration and size distribution using nanoparticle tracking analysis (NTA).
Immunoassay: Detects exosome-specific markers like CD9, CD63, CD81, and Alix.

2. Additional EV characterization

Transmission Electron Microscopy (TEM): Visualizes the morphology of exosomes.
Flow Cytometry: Analyzes surface markers on exosomes.
Fluorescent nanoparticle tracking analysis (NTA): Fluorescence-based detection of specific biomarkers.
Dynamic light scattering (DLS): Size distribution and polydispersity in a suspension
Mass spectrometry: Determine the density of the exosomes
RNA sequencing (RNA-seq): Analyzes exosomes RNA content, allowing for the identification and quantification of exosomal RNA species.
