Multi-Species Native Hemoglobin Reagents

Introduction to Hemoglobin

Hemoglobin (Hgb) is a complex iron-containing protein found in erythrocytes (red blood cells) of most vertebrates that plays a vital role in carrying oxygen and carbon dioxide within the blood.

It is also the component that makes our blood red. It is found in animals containing red-colored blood i.e. in almost every vertebrate except fishes of the Channichthyidae family and in a few invertebrates. Beside RBC, hemoglobin is also present in alveolar cells, macrophages, some neurons of the midbrain, mesangial cells of kidneys, hepatocytes, vaginal epithelial and cervical cells, etc.

Molecular Structure and Function

Hgb is a globular metalloprotein with a quaternary structure. One hemoglobin molecule is composed of four subunits, each containing one polypeptide chain (a globin protein) attached to a heme prosthetic group. Each subunit weighs about 16,000 Dalton, making the total molecular weight of a Hgb molecule of 64,000 Dalton.

The polypeptide chains in adults are of two types, the alpha chain and the beta chain containing 141 and 146 amino acids respectively. Adult hemoglobin has two alpha subunits (α1 and α2) and two beta subunits (β1 and β2), combined as two αβ dimers arranged around a 2-fold axis of symmetry. Each subunit has a heme attached to the globin protein.

The heme group contains an iron (ferrous ion, Fe2+) held in the center of a porphyrin ring binding with the nitrogen atoms of the ring. Each Fe2+ ion can bind with one oxygen (O2) molecule or one carbon dioxide (CO2) molecule.

The primary function of hemoglobin is to carry oxygen and carbon dioxide gases. It also serves the secondary function of maintaining blood pH and buffering the blood.

Clinical Significance: Anemia, Diabetes, and Veterinary Health

Hemoglobin testing plays a critical role in both human and veterinary diagnostics. A standard hemoglobin blood test is widely used to evaluate oxygen-carrying capacity and serves as a primary screening tool for anemia, hemorrhage, and other hematological disorders. In metabolic disease monitoring, Hemoglobin A1c (HbA1c) is considered the gold-standard biomarker for assessing long-term blood glucose control in diabetic patients by reflecting average glucose levels over the previous two to three months.

In veterinary medicine, species-specific hemoglobin analysis is valuable for diagnosing immune-mediated hemolytic anemia in dogs, monitoring exercise physiology and performance in horses, and evaluating health status in livestock and laboratory animals. Hemoglobin antigens are also widely used in immunoassay development, antibody production, and comparative biomedical research.
