Antibody-drug conjugates (ADCs) represent a revolutionary class of targeted therapeutics that combine the specificity of monoclonal antibodies with the cytotoxic potency of small-molecule payloads. A critical step in the ADC mechanism of action is antibody internalization—the process by which the antibody binds to its target antigen on the cell surface, undergoes receptor-mediated endocytosis, and is trafficked into intracellular compartments such as endosomes and lysosomes. Effective internalization is essential for intracellular payload release and cytotoxicity. To accelerate ADC development, accurate, sensitive, and scalable internalization assays are indispensable for screening, optimizing, and validating antibody candidates.
Our Antibody Internalization Assay Reagents are designed to streamline this process with high sensitivity and flexibility. We offer three robust tools tailored for different research needs: the DT3C IgG Labeling Reagent, the MMAE IgG Labeling Reagent, and the pH-Sensitive IgG Labeling Reagent. These reagents enable direct labeling of antibodies without requiring complex conjugation protocols or advanced equipment, making them ideal for rapid evaluation of internalization behavior and intracellular payload delivery potential.
pH-Sensitive IgG Labeling Reagent: It is a fluorogenic dye system that becomes highly fluorescent in acidic environments such as endosomes and lysosomes. The reagent uses a pH-sensitive fluorescent dye conjugated Fc binding protein, which forms a complex with IgG antibodies. Upon internalization of the antibody-reagent complex into acidic compartments, the fluorescence signal increases dramatically. By quantifying the signal strength, researchers can evaluate the efficiency and kinetics of internalization, gaining insights into antibody uptake mechanisms, trafficking behavior, and therapeutic potential.
Payload-Conjugated IgG labeling Reagent: It simulates the behavior of true ADCs by attaching cytotoxic payloads, such as DT3C or VcMMAE, directly to antibodies, enabling simultaneous assessment of antibody internalization and cytotoxicity. These reagents utilize a payload-conjugated Fc-binding protein to label IgG antibodies from various species, forming antibody–payload complexes. Upon binding to target cells and subsequent internalization, the payload is released inside the cell, leading to cell death. By measuring cell viability—typically using a CCK-8 assay—researchers can evaluate both the internalization efficiency and functional potency of candidate monoclonal antibodies. This approach offers a simple yet biologically relevant platform, making it ideal for the high-throughput screening and early-stage optimization of ADC candidates.
