Liraglutide is a human glucagon like peptide-1 (GLP-1) receptor agonist. The Liraglutide High Sensitivity ELISA Kit employs a sandwich enzyme-linked immunosorbent assay (ELISA) format, widely regarded as the gold standard for quantitative protein detection. This method utilizes two highly specific antibodies that recognize distinct epitopes on the liraglutide molecule, ensuring exceptional specificity and minimizing cross-reactivity with other GLP-1 analogs or endogenous peptides.

The assay workflow begins with pre-coated capture antibodies immobilized on the surface of 96-well microplate wells. When samples or standards are added, liraglutide molecules in the sample bind specifically to these capture antibodies. After a washing step to remove unbound components, a biotinylated detection antibody is introduced, forming a sandwich complex with the captured liraglutide. Streptavidin-horseradish peroxidase (HRP) conjugate is then added, which binds to the biotinylated detection antibody, creating an enzyme-labeled complex .
The final step involves adding a chromogenic substrate that undergoes enzymatic conversion by HRP, producing a colored reaction product. The prolonged action of liraglutide (as compared to endogenous GLP-1) is achieved by attaching a fatty acid molecule at one position of the GLP-1-(7-37) molecule, enabling it to both self-associate and bind to albumin within the subcutaneous tissue and bloodstream. The active GLP-1 is then released from albumin at a slow, consistent rate. |
