High-Sensitivity Measles IgG ELISA Kit for Quantitative Detection

The Measles IgG ELISA Kit is a highly reliable and efficient in vitro diagnostic tool specifically developed for the quantitative detection of Measles virus IgG antibodies in human serum, plasma, and other relevant biological samples. This kit utilizes the enzyme - linked immunosorbent assay (ELISA) technique, which is widely recognized for its high sensitivity, specificity, and accuracy in immunological testing. It plays a crucial role in various fields such as clinical diagnosis, epidemiological surveillance, and vaccine efficacy evaluation.

Pathogens of Measles

Measles is caused by the measles virus, which belongs to the Paramyxoviridae family and the Morbillivirus genus. It is an enveloped single stranded negative stranded RNA virus. The virus surface contains multiple key proteins, among which hemagglutinin (H protein) and fusion protein (F protein) are the key factors for its infection and pathogenicity. H protein is responsible for binding to receptors (such as CD150/SLAM and nectin-4) on the surface of host cells (such as lymphocytes and epithelial cells), while F protein mediates the fusion of viral envelope and cell membrane, allowing viral RNA to enter the cytoplasm for replication. These proteins are also the main targets for the human immune system to recognize and produce neutralizing antibodies.

Epidemiology and infectivity

Measles is one of the most contagious diseases known. The basic reproduction number is as high as 12-18, which means that a case can be transmitted to 12 to 18 people in a completely susceptible population.

Background introduction and importance

Measles is a highly contagious acute respiratory disease caused by the measles virus, mainly transmitted through droplets. Despite the existence of safe and effective vaccines, measles remains one of the leading causes of child mortality worldwide, especially in areas with low vaccination rates. The measles virus IgG antibody is a key indicator of adaptive immune response. After acute infection, IgM antibodies first appear, followed by the production and sustained presence of IgG antibodies, which typically provide lifelong immunity. The quantitative detection of measles IgG antibodies has multiple important significance:

Key Components & Specifications

Performance Characteristics

  • Analytical Sensitivity: <10 mIU/mL (the lowest concentration that can be reliably detected).
  • Measuring Range: 10-2000 mIU/mL (covering a wide range from negative to high titers).
  • Precision: The coefficient of variation (CV) for intra assay and inter assay is both less than 10%.
  • Specificity: No cross reactivity with other common pathogens such as rubella virus, mumps virus, and parainfluenza virus.
  • Accuracy: Good correlation with international standards or other recognized reference methods (R ²>0.98).

Sample Processing Requirements

  • Sample type: human serum or heparin/EDTA anticoagulant plasma.
  • Collection: Use standard venous blood collection procedures. Avoid using samples that are hemolytic, lipophilic, or heavily contaminated.
  • Processing: Whole blood samples should be naturally clotted at room temperature, and serum should be separated by centrifugation (e.g. 1000 × g, 10 minutes). Plasma samples need to be separated by centrifugation.
  • Storage: If the separated sample is used within 24 hours, it can be stored at 2-8 ° C. If long-term storage is required, it should be divided and frozen at -20 ° C or lower. Avoid repeated freeze-thaw cycles (more than 3 times).

Data Calculation

  • Calculate the average OD value: After deducting the blank hole OD value, calculate the dual hole average OD value of the calibration sample and sample.
  • Draw a standard curve: Using the known concentration of the calibration standard as the X-axis (logarithmic coordinate), modern enzyme-linked immunosorbent assay software can usually automatically complete this process.
  • Interpolation calculation: Interpolate the corresponding antibody concentration from the standard curve based on the average OD value of the sample.

Precautions

  • For Research Use Only (RUO): This product is not intended for clinical diagnostic procedures unless permitted by local regulations.
  • Biosafety: All human samples should be considered potentially infectious and handled in accordance with Biosafety Level 2 (BSL-2) operating procedures.
  • Reagent: The termination solution contains dilute sulfuric acid, which is corrosive. If in contact with skin or eyes, please rinse immediately with plenty of water.
  • Quality control: Each experiment must include calibration curves and negative/positive quality control samples. The quality control product results must be within the expected range for the experiment to be effective.

Applications of the Measles IgG ELISA Kit

Assessment of Post-Vaccination Immunity

Assessment of Post-Vaccination Immunity: The measles vaccine is highly effective, but not all individuals develop a protective immune response after vaccination. Furthermore, immunity can wane over time, especially in individuals who received a single dose. This kit allows for the quantitative assessment of antibody titers in vaccinated individuals. A titer above a certain threshold (e.g., 200 mIU/mL) is generally considered protective.

Confirmation of Past Measles Infection

Confirmation of Past Measles Infection

In cases where a patient presents with symptoms suggestive of measles but a clinical diagnosis is uncertain, the presence of a high titer of measles IgG antibodies, especially when a paired sample shows a significant rise in titer, can confirm a past infection. It's important to note that IgM antibodies are typically used for acute infection diagnosis, while IgG is a marker of long-term immunity.

ParameterDetails
Product nameMeasles IgG ELISA Kit
SampleSerum, Plasma and Cerebrospinal Fluid.
Intended UseThe Measles Virus IgG tests are quantitative and qualitative immunoassays for the detection of human antibodies in serum or plasma directed against Measles Virus. The Measles Virus IgG assay is recommended for determination of immune status and for detection of intrathecal synthesized IgG antibodies in cerebrospinal fluid.
Performance CharacteristicsTo evaluate the performance characteristics of the Measles Virus IgG, 195 serum samples from seronegative donors, patient samples, sera from children and unselected samples were examined in comparison to Measles virus IgG ELISA test system from two other manufacturer. Analyses have been performed according to the corresponding instructions for use. Borderline results were not included in the calculation of sensitivity and specificity.
StorageMeasles Virus, in common with Mumps virus, Parainfluenza Viruses and RS Virus belongs to the family of paramyxoviridae and has a worldwide distribution. According to WHO estimates, one million individuals die as a consequence of Measles Virus infections every year. A number of countries, along with the WHO, had set themselves the goal of eradicating measles by a program of vaccination.

Conclusions

The Measles IgG ELISA Kit is an invaluable tool for both clinical diagnostics and public health surveillance. Its ability to provide a precise, quantitative measure of immunity enables healthcare providers and public health officials to make informed decisions regarding vaccination strategies, outbreak management, and individual patient care. By utilizing this advanced serological method, we can continue to advance our efforts toward the global eradication of measles.

Please refer to the instruction manual, which usually provides conversion formulas or conversion factors. If not, please contact technical support.

It is usually recommended to have an initial dilution ratio of 1:101 to minimize sample matrix effects and ensure that the OD values of the vast majority of samples fall within the linear range of the standard curve. For samples with extremely high titers (OD values exceeding the highest standard), they should be further diluted and retested.

Check if the calibration material is prepared incorrectly, if there are bubbles in the well, if the enzyme-linked immunosorbent assay (ELISA) is functioning properly, or if the incubation time/temperature is accurate. Suggest preparing calibration samples again and running the standard curve.
