Post-translational modifications (PTMs) are essential processes that regulate protein structure, function, and stability. Carboxylation is one of PTMs that involves the addition of a carboxyl group to specific amino acid residues, most commonly glutamate (Glu), resulting in the formation of γ-carboxyglutamate (Gla) residues. This modification is catalyzed by the enzyme γ-glutamyl carboxylase, with vitamin K serving as a critical cofactor in the process.
Carboxylation plays a pivotal role in various biological pathways, including blood clotting, bone growth, and extraosseous calcification. It is primarily observed in proteins involved in the blood clotting cascade, such as factors II, VII, IX, and X, protein C, and protein S. Additionally, carboxylation is crucial for the regulation of bone growth and the prevention of abnormal calcification.

Carboxylation is catalyzed by the enzyme γ-glutamyl carboxylase, which is localized in the endoplasmic reticulum. The process of this modification requires several essential components and follows a series of steps:
γ-Glutamyl carboxylated proteins can be classified into three main groups based on their structural and functional characteristics:
Some important aspects of carboxylation in biological processes include:
Understanding the importance of protein carboxylation in biological processes has implications for both basic research and clinical applications. It provides insights into the molecular mechanisms underlying various physiological functions and helps uncover the role of carboxylation in disease development and progression. Furthermore, targeting protein carboxylation pathways may offer therapeutic opportunities for the treatment of related disorders.
