What is RPPA?

RPPA technology is a high-throughput proteomics technology that combines planar high-precision large-scale sample protein antigen microarray printing and antibody detection.

The RPPA technology platform is rigorously designed, the process is highly automated, and the quality control is extremely strict. Each time the sample protein antigen microarray is printed, multiple positive standard cell line samples are arrayed together with the sample to be analyzed to ensure the high traceability, high credibility and repeatability of the experimental results. RPPA can analyze up to 500 or more proteins of different abundance in 15 mg of tissue (rice grain size) at one time, and the antibody reaction process and target signal acquisition are independent of each other, avoiding the crosstalk between different target antibody reactions and the incompatibility of signal acquisition caused by the large difference in abundance of different targets. It has ultra-high specificity and sensitivity that other high-throughput proteomics cannot match. It can perform systematic and in-depth detection of a large number of representative targets such as cell surface receptor proteins, key cell signaling proteins and protein modifications (phosphorylation, acetylation, methylation, etc.), proteases, transcription factors, etc., and can also customize the target protein to be studied according to needs for analysis, including direct and indirect upstream and downstream protein network analysis. RPPA can be applied to different fields such as protein function and regulatory mechanism analysis, panoramic scanning of cell signal transduction, tumor marker and protein molecular typing, drug target discovery and drug mechanism analysis.

Service Advantages

RPPA Service Advantages-1

Small Sample Requirement

20-40 micrograms of protein can achieve 380 target detection, which can meet the wide range of samples analyzed by RPPA, including clinical solid tumor surgical samples, puncture biopsy specimens, PDX model samples, in vitro cultured cells, 3D cultures, etc.

Standardized Sample Processing and Analysis

Proteins and protein modification sites are detected in a linear state after protein denaturation, with high sample stability, eliminating potential problems in epitope recognition. Based on the standardized technical process of the IS015189 quality management system, with strict quality control, it can efficiently perform parallel detection, data analysis and evaluation of large samples.
RPPA Service Advantages-2

Target Independent Detection High Sample Throughput

For one detection target, 100-1000 samples are analyzed in parallel under the same reaction microenvironment. The sample throughput is significantly higher than other proteomics technology platforms, and each target detection reaction is performed independently, completely eliminating the problem of signal crosstalk between different targets.

High Compatibility of Target Signals with Different Abundances

The sample microarray has been carefully designed, and strict and comprehensive controls for multiple cell line samples have been established. Each sample (including the sample to be tested and the control sample) is serially diluted, and the target signal is relatively quantified, which can take into account the target signals with different abundances, and can detect small signal differences and low-abundance signals with high sensitivity.
