IRF5 Introduction
IRF5, or interferon regulatory factor 5, is a transcription factor belonging to the IRF family, which plays a central role in regulating innate and adaptive immune responses. It is activated downstream of pattern-recognition receptors such as Toll-like receptors and cytosolic nucleic acid sensors. Once activated, IRF5 translocates to the nucleus, where it binds specific DNA motifs to induce the expression of type I interferons and pro-inflammatory cytokines. IRF5 is predominantly expressed in immune cells including monocytes, macrophages, dendritic cells, and B cells, positioning it as a key regulator of inflammation and antiviral defense.

Functionally, IRF5 orchestrates the transcription of numerous genes involved in immune activation, shaping Th1 and Th17 immune responses and influencing macrophage polarization toward a pro-inflammatory M1 phenotype. It modulates signaling pathways such as NF-κB and JAK–STAT, amplifying cytokine production and promoting efficient pathogen clearance. IRF5 is also involved in apoptosis regulation and cellular stress responses, linking inflammation with broader immune and metabolic networks. Experimental studies have shown that IRF5 deficiency reduces inflammatory cytokine expression and alters macrophage behavior, while pathogenic hyperactivation leads to excessive immune stimulation.
Clinically, IRF5 is strongly associated with autoimmune and inflammatory diseases. Polymorphisms within the IRF5 gene are among the most consistent genetic risk factors for systemic lupus erythematosus and are also linked to rheumatoid arthritis, inflammatory bowel disease, Sjögren's syndrome, and systemic sclerosis. Dysregulated IRF5 signaling contributes to chronic inflammation, aberrant interferon responses, and tissue damage characteristic of these conditions. Beyond autoimmunity, IRF5 has emerging relevance in cancer biology, where its role in shaping the immune microenvironment may influence tumor progression and therapy response. These disease associations highlight IRF5 as a crucial immune regulator and a promising biomarker and therapeutic target in immunology and precision medicine.
