BPNT1 Full Name
3(2), 5-bisphosphate nucleotidase 1
BPNT1 Introduction
Bisphosphate 3′-nucleotidase 1 (BPNT1) is a phosphatase that hydrolyzes 3′-phosphoadenosine 5′-phosphate (PAP), a toxic byproduct of sulfation reactions that must be cleared to allow continued activity of sulfotransferases. The enzyme localizes to the Golgi lumen through a cleavable N-terminal signal sequence and is essential for sulfate metabolism across virtually all tissues. The BPNT1 gene maps to chromosome 1q41 and gives rise to a single highly conserved ~33 kDa protein.

PAP Catabolism and Sulfation Pathway Maintenance
BPNT1 acts as the final step in the sulfate activation pathway, hydrolyzing PAP into 5′-AMP and inorganic phosphate. PAP is generated at every sulfotransferase reaction cycle and acts as a competitive inhibitor of these enzymes, so its removal by BPNT1 is essential to sustain sulfation of hormones, neurotransmitters, proteoglycans, and xenobiotics. Loss of BPNT1 causes PAP accumulation, inhibition of sulfotransferases, and disruption of Golgi sulfation reactions, with downstream effects on extracellular matrix composition, drug metabolism, and hormone bioactivity.
Metabolic and Inflammatory Disease
Recent studies have linked BPNT1 activity to the regulation of brown adipose tissue thermogenesis and to obesity, with BPNT1-deficient mice displaying increased energy expenditure and protection from diet-induced weight gain. In cancer, BPNT1 supports sulfation of proteoglycans in the tumor microenvironment and may influence sensitivity to heparan-sulfate-dependent signaling pathways. BPNT1 inhibitors are also being explored as adjuncts to enhance the efficacy of sulfated drug substrates, including certain steroid hormones and catecholamines used in clinical practice.
