Staphylococcus aureus enterotoxins B (SEB)

Staphylococcus aureus is a spherical, Gram-positive coccus of approximately 1 um in diameter and forms short chains or grape-like clusters. Several virulence factors of S. aureus strains have been reported, and enterotoxins are the most notable virulence factors, as they can cause the sporadic food-poisoning syndromes or foodborne outbreaks.

22 different staphylococcal enterotoxins (SEs) have been described since 1959, including enterotoxins A (SEA), B (SEB), C (SEC1), C2 (SEC2), C3 (SEC3), D (SED), and E (SEE). These toxins are non-glycosylated, globular single-chain proteins with low molecular weight ranging from 19 to 29 kDa. SEB is one of the most common biotoxins reported and a typical biotoxin that causes food poisoning.

Creative Diagnostics has developed a range of SEB antigens and antibodies that can be used for in vitro detection and diagnosis research. They are well-validated in double-antibody sandwich enzyme-linked immunosorbent assay.

Pathogenicity

SEB, one of the most common and potent superantigens of Staphylococcus aureus, binds to the α chain of MHC II and the specific vβ region of the T cell receptor (TCR) to form a terpolymer, resulting in massive proliferation and activation of monocyte-macrophages and T cells. And induce high levels of proinflammatory cytokines and chemokines, which may lead to toxic shock syndrome (TSS) in patients. SEB is closely related to food poisoning and can cause symptoms such as vomiting, abdominal pain and diarrhea.

Detection

SEB has the characteristics of high temperature resistance, acid and alkali resistance, easy to produce aerosols, etc., and has been listed as a prohibited warfare agent by the the Biological Weapons Convention. Therefore, the study of the highly sensitive and rapid detection method of SEB is of great significance for the clinical diagnosis, prevention and control of food poisoning.

Immunoassay is mainly based on the principle of specific binding of antigen and antibody, including immunoagglutination assay, agarose diffusion method and enzyme-linked immunosorbent assay (ELISA). Sandwich ELISA and icELISA are commonly used to detect SEB, with the former having higher sensitivity. At present, commercial ELISA kits have been developed.
