Secondary VHH Antibodies

Overview

VHH is the variable domain of camelids heavy chain antibodies (HCAb), and it also called single-domain antibody (sdAb) or nanobody. Due to the various natural features, VHH is widely considered to have a bright future in the field of replacing traditional antibodies. At present, as an excellent and sustainable alternative tool, secondary VHH antibody have been widely used, which can effectively solve pain point problems of traditional polyclonal secondary antibodies.

Advantages of VHH

The Fig.1 shows the structural difference between the VHH and the conventional IgG. Camelid HCAb is charactered by the absence of light chains and the lack of the CH1 in the heavy-chain, and it contains only two constant regions, CH2 and CH3, and a variable region, VHH, in each chain.

With a longer CDR3 region, VHH has the ability to bind to a wide range of antigen epitopes. It is also more hydrophilic and stable, and can withstand high temperatures and extreme pH conditions. VHH also shows excellent tissue penetration due to the smallest size which allows them to move freely through the tissue. In addition, VHH can be easily genetically engineered at very low cost, which undoubtedly makes VHH-related products highly commercial value.

Innovatively Secondary VHH Antibody

Conventional polyclonal anti-IgG/IgM secondary antibodies are essential tools for many molecular biology techniques and diagnostic tests. However, the large size of conventional secondary antibodies (∼10–15 nm) limits tissue penetration and introduces considerable label displacement, reducing the obtainable image resolution by super resolution fluorescence microscopy methods. Also, laboriously quality control is required between different batches. In addition, animal-based production leads to the killing of many animals such as goats, sheep or rabbits, which is a major animal welfare issue.

VHHs/SdAbs may be the best sustainable candidates to replace such reagents. In addition to excellent antigen specificity and reproducible production, VHHs/SdAbs are also particularly useful for super resolution imaging. Site-specifically labeled SdAbs can place fluorophores closer than 2 nm to the antigen and, despite their small size, even tolerate up to three dyes. In general, VHHs/SdAbs have been shown to have excellent ability in many applications, such as ELISA, WB, IF, etc.
