Introduction
Staphylococcus aureus Clumping Factor A (ClfA) is a surface-exposed virulence protein of S. aureus, belonging to the MSCRAMM adhesin family. It mediates bacterial attachment to host tissues by binding fibrinogen via a "dock, lock, latch" mechanism, crucial for immune evasion and biofilm formation. ClfA facilitates bloodstream infections, endocarditis, and medical device colonization by enabling bacterial clumping under physiological shear stress. Its fibrinogen-binding domain (residues 221–550) is a prime target for therapeutic antibodies and vaccines against antibiotic-resistant strains, including MRSA.
