Protein ubiquitination is a versatile post-translational modification (PTM) that regulates various fundamental characteristics of proteins, including stability, activity, and localization. By connecting ubiquitin proteins at specific locations, this modification plays a crucial role in cell signaling, protein degradation, and maintenance of cellular homeostasis. Dysregulation of protein ubiquitination has been implicated in the pathogenesis of numerous diseases, including cancer, neurodegenerative disorders, and immune-related disorders.

The process of protein ubiquitination can be further extended by the addition of additional ubiquitin molecules to the initial ubiquitin modification. This leads to the formation of polyubiquitin chains, where individual ubiquitin molecules are linked together through specific lysine residues on ubiquitin itself. The type of linkage between ubiquitin molecules in the chain, such as K48 or K63 linkages, determines the fate and function of the modified protein.
The fate of ubiquitinated proteins is not solely determined by the addition of ubiquitin molecules. Deubiquitinating enzymes (DUBs) play a crucial role in the removal of ubiquitin from target proteins, thereby reversing the ubiquitination process. DUBs can cleave the isopeptide bond between ubiquitin and the target protein, allowing for the recycling of ubiquitin and the restoration of the unmodified protein.
