Intended Use

ITGA2B/ITGB3 (Human) ELISA Kit is a sandwich enzyme immunoassay for the quantitative measurement of human ITGA2B/ITGB3.

Contents of Kit

1. Human GPIIb/IIIa Microplate: A 96-well polystyrene microplate coated with a polyclonal antibody against GPIIb/IIIa. 96 (8×12) wells
2. Sealing Tapes: Pre-cut, pressure-sensitive sealing tapes that can be cut to fit the format of the individual assay. 3 slices
3. Human GPIIb/IIIa Standard: Human platelet GPIIb/IIIa in a buffered protein base (lyophilized). 60 ng
4. Biotinylated Human GPIIb/IIIa Antibody (100×): A 100-fold concentrated biotinylated polyclonal antibody against human GPIIb/IIIa. 80 μl
5. MIX Diluent Concentrate (10×): A 10-fold concentrated buffered protein base. 30 ml
6. Wash Buffer Concentrate (20×): A 20-fold concentrated buffered surfactant. 30 ml × 2
7. Streptavidin-Peroxidase Conjugate (SP Conjugate): A 100-fold concentrate. 80 μl
8. Chromogen Substrate: A ready-to-use stabilized peroxidase chromogen substrate tetramethylbenzidine. 8 ml
9. Stop Solution: A 0.5 N hydrochloric acid to stop the chromogen substrate reaction. 12 ml

Storage

1. Upon arrival, immediately store components of the kit at recommended temperatures up to the expiration date.
2. Store SP Conjugate and Biotinylated Antibody at -20°C.
3. Store Microplate, Diluent Concentrate (10×), Wash Buffer, Stop Solution, and Chromogen Substrate at 2-8°C.
4. Unused microplate wells may be returned to the foil pouch with the desiccant packs and resealed. May be stored for up to 30 days in a vacuum desiccator.
5. Diluent (1×) may be stored for up to 30 days at 2-8°C.
6. Store Standard at 2-8°C before reconstituting with Diluent and at -20°C after reconstituting with Diluent.

Precision

Intra-assay and inter-assay coefficients of variation were 4.9% and 7.6% respectively.

Sensitivity

The minimum detectable dose of GPIIb/IIIa is typically ~ 1 ng/ml.

General Description

Platelet membrane glycoprotein IIb/IIIa (GPIIb/IIIa, integrin αIIbβ3) is a member of the integrin family of cell membrane receptors that play key roles in thrombus formation, platelet aggregation, embryogenesis, and intercellular adhesion. Each integrin receptor complex consists of a heavy (alpha) and a light (beta) chain associated as a calcium-dependent heterodimer with a molecular mass of 140 kDa and 90 kDa respectively. GPIIb/IIIa serves as an inducible receptor for fibrinogen, fibronectin, von Willebrand factor, and vitronectin. The simultaneous occupancy on adjacent platelets of receptors with dimeric fibrinogen molecules leads to platelet aggregation. Hereditary defects of the GPIIb/IIIa receptor cause Glanzmann's thrombasthenia (GT), an autosomal recessive bleeding disorder.
