Intended Use
For the simultaneous quantitative determination of multiple human biomarker concentrations in cell culture supernates, serum, plasma, and platelet-poor plasma.
Contents of Kit
1. Standard Cocktail 1. 2 vials of recombinant human cytokines in a buffered protein base with preservatives, lyophilized. Discard after use. Use a fresh standard and control for each assay.

6. Human Performance Panel Biotin-Antibody Cocktail. 1 vial of a concentrated biotinylated antibody cocktail, lyophilized. STORAGE OF OPENED, DILUTED, OR RECONSTITUTED MATERIAL: May be stored for up to 1 month at 2-8°C.* Once diluted, 1× solutions must be discarded. Use fresh diluents for each assay.
7. Streptavidin-PE. 0.250 mL of a concentrated streptavidin-phycoerythrin conjugate with preservatives. STORAGE OF OPENED, DILUTED, OR RECONSTITUTED MATERIAL: May be stored for up to 1 month at 2-8°C.* Once diluted, 1× solutions must be discarded. Use fresh diluents for each assay.
8. Micropartice Diluent. 6 mL of a buffered protein base with blue dye and a preservative. STORAGE OF OPENED, DILUTED, OR RECONSTITUTED MATERIAL: May be stored for up to 1 month at 2-8 °C.*
9. Biotin Antibody Diluent 2. 8 mL of a buffered protein base with preservative. STORAGE OF OPENED, DILUTED, OR RECONSTITUTED MATERIAL: May be stored for up to 1 month at 2-8 °C.*
10. Calibrator Diluent. 21 mL of a buffered protein base with preservatives. Use diluted 1:2 for cell culture supernate samples. Use undiluted for serum/plasma samples. STORAGE OF OPENED, DILUTED, OR RECONSTITUTED MATERIAL: May be stored for up to 1 month at 2-8 °C.*
11. Wash Buffer Concentrate. 21 mL of a 25-fold concentrated solution of buffered surfactant with preservative. May turn yellow over time. STORAGE OF OPENED, DILUTED, OR RECONSTITUTED MATERIAL: May be stored for up to 1 month at 2-8 °C.*
12. Microplate. 1 flat-bottomed 96-well microplate used as a vessel for the assay.

General Description
Immune-based therapies have become promising approaches for the treatment of cancers. Cytokine-release syndrome (CRS), associated with some cancer immunotherapies, can be fatal if not properly identiffed and managed. Monitoring circulating levels of inflammatory biomarkers is an important step in determining the response to immunotherapy treatment and in predicting the likelihood of adverse effects like CRS.
Assessing the levels of multiple biomarkers may be more revealing than analyzing a single protein. Quantifying multiple biomarkers on an individual level using enzyme-linked immunosorbent assays (ELISA), for example, can be time consuming and expensive. Multiplex assays allow measurement of multiple biomarkers simultaneously and conserves precious samples by using a small sample volume.
The Immunotherapy Performance Assay is a multiplex Luminex bead-based assay that allows for rapid, sensitive, and economical proffling of biomarkers associated with the host response to immune-based therapies.
