A class of viruses named herpesviruses causes multiple human infections. Herpesviruses establish latent infections that can reactivate and cause diverse clinical symptoms.
Herpesviruses gain entry into cells through coordinated interactions between several glycoproteins present on their surface. The virus attaches to the host cell initially which anchors it without activating entry into the cell. The virus attaches to host cells through interactions between multiple viral glycoproteins and different binding receptors. Herpesviruses use multifunctional viral glycoproteins to perform both receptor binding and membrane fusion actions. The current model demonstrates that entry receptor interaction initiates structural changes in viral glycoproteins which activate gB to perform membrane fusion.

The Herpes Simplex Virus Type 1 uses gD as the protein responsible for binding to receptors. The receptor-bound protein connects with gH-gL which results in activation of gB to initiate fusion.
EBV entry into B cells operates through a similar mechanism but employs gp42 as its receptor-binding protein.
HCMV receptor binding is mediated by two distinct complexes: HCMV receptor binding happens through two distinct protein complexes: gO with gH-gL forming a trimeric complex and UL128 along with UL130 and UL131A combined with gH-gL creating a pentameric complex. The virus infects the cell after its membrane joins with the host cell membrane through these interactions and structural changes.
