Neutralizing Neuropilin-1 (NRP1) antibodies: Trusted Tools for SARS-CoV-2 Intervention Research

Neuropilin-1 (NRP1) is a transmembrane glycoprotein that functions as coreceptors for VEGF. It exhibits high expression in dendritic cells, where it mediates DC-induced T cell proliferation, and in respiratory / olfactory epithelium cells. The primary structure of NRPI is well conserved and is divided into four domains, CUB (a1/a2) domain, FV/FVIII (b1/b2) domain, MAM (c) domain, and (d) domain.

Multiple studies reveal that the NRP1 expressed in the respiratory and olfactory epithelial facilitates SARS-CoV-2 entry into the host cell.

1) SARS-CoV-2 uses Spike (S) protein for host cell attachment and entry.

2) The host protease furin cleaves the full-length precursor S glycoprotein into two associated polypeptides: S1 and S2.

3) Cleavage of S generates a polybasic Arg-Arg-Ala-Arg C-terminal sequence on S1 fragment.

4) Modified S1 fragment binds directly to the NRPI b1 domain of cell surface NRP1 and significantly potentiates viral infectivity.

Thus, RNAi, selective inhibitors, and antibodies could all exert the blockage. Cantuti-Castlelvetri revealed this antibody-mediated NRP1-Spike blocking interaction reduces viral infectivity in vitro and makes it a potential therapeutic target for antiviral intervention. This specific blockage also provide a route for COVID-19 therapies.
