What is Single-domain Antibody?

A Single-domain Antibody (sdAb), also called nanobody, is a natural single monomeric variable antibody domain found in heavy-chain-only antibodies isolated from camelids (llama, alpaca). A single-domain antibody is structurally different from conventional antibodies, which are heterodimeric in nature containing two heavy and two light chains. These antibodies have an advantage of small size of about 12–15 kDa.

In this case, in order to verify the affinity of the developed single-domain antibody, Creative Diagnostics used BLI (Bio-Layer Interferometry) technology for binding kinetics testing. The figure below shows a typical BLI sensogram, where the horizontal axis is time (seconds) and the vertical axis is the binding response signal (nm offset). The blue curve intuitively shows the antibody's quick elevation in the binding phase and slow decay in the dissociation phase, and the gray dotted line in the middle represents the demarcation between the two phases. The "fast binding, slow dissociation" property indicates that the antibody has high affinity and good stability to the target antigen.
