What is Single-domain Antibody?

A Single-domain Antibody (sdAb), also called nanobody, is a natural single monomeric variable antibody domain found in heavy-chain-only antibodies isolated from camelids (llama, alpaca). A single-domain antibody is structurally different from conventional antibodies, which are heterodimeric in nature containing two heavy and two light chains. These antibodies have an advantage of small size of about 12–15 kDa.
Single-domain antibodies (sdAbs) are highly specific and retain high affinity to their target antigen. They can bind to hidden or cryptic epitopes that are inaccessible to conventional antibodies. They are extremely stable against chemical and thermal denaturation. They can also be easily expressed recombinantly, because of their small size and simple structure.
