Anti-Human IgG Fab polyclonal antibody [CL-MMAE] is a secondary antibody-drug conjugates for pre-screening antibodies with a human Fab moiety to determine their cytoxocity as MMAE bioconjugates. It alone displays no obvious toxicity against multiple cell lines at 2 ug/ml (13 nM) or lower concentration. When applied in combination with tumor specific monoclonal antibodies, it can help determine the cytotoxic potential for these antibodies against target cell lines.

General Notes

SecADC™ Anti-Human IgG Fab polyclonal antibody [CL-MMAE] can be used in cell-based cytotoxicity assays to pre-screening monoclonal antibodies as ADC candidates against tumor cells. This product is anti-human IgG Fab specific antibody conjugated to monomethyl auristatin E (MMAE) with a cleavable linker. The antibody portion is a polyclonal antibody (full length IgG) which is specific to the Fab region of human IgGs. Monomethyl auristatin E (MMAE) is a cytotoxic small molecule which inhibits cell division by blocking the polymerization of tubulin. The cleavable linker connecting MMAE to the antibody is stable in extracellular fluid, but is cleaved by cathepsin in endosome once the conjugate has entered a cell via endocytosis.
