A DNA sequence encoding the extracellular domain (Met1-Ala105) of human CD3D (P04234-1) was fused with the C-terminal his-tagged Fc region of human IgG1 at the C-terminus, constructed the plasmid 1; A DNA sequence encoding the extracellular domain (Met1-Asp126) of human CD3E (P07766-1) was fused with the C-terminal flag-tagged Fc region of human IgG1 at the C-terminus, constructed the plasmid 2. The two plasmids were co-expressed and the human CD3D/CD3E heterodimer was purified.

Molecular Weight
The recombinant heterodimer of human CD3D/CD3E consists of 687 (334+353) amino acids and predicts a molecular mass of 77.5 (37.7+39.8) kDa. It migrates as an approximately 44-54 KDa band in SDS-PAGE under reducing conditions due to glycosylation.

Introduction
CD3D and CD3E are two chains that are part of the CD3 complex, a crucial component of the T-cell receptor (TCR) complex on T lymphocytes. This complex plays a key role in signaling pathways within T cells, particularly when they recognize antigens. CD3D and CD3E are single-pass type I membrane proteins that, along with other CD3 chains (CD3G and CD3Z), help transmit signals from the TCR to the cell's interior.
