General Notes
Clone WH9 is a monoclonal antibody raised against Der p 7, a major group 7 allergen of the house dust mite Dermatophagoides pteronyssinus. WH9 has been shown to inhibit human IgE binding to Der p 7 by up to 60%, making it a valuable tool for studying allergen‑antibody interactions relevant to allergic disease. The epitope recognized by WH9 on Der p 7 is conformational (loop‑like) and comprises four critical residues: Serine 156, Leucine 158, Aspartic acid 159, and Proline 160. Molecular docking and homology modeling of the Der p 7‑WH9 complex revealed seven hydrogen bonds, two electrostatic interactions, and four hydrophobic interactions between the antibody and the allergen. WH9 exhibits cross‑reactivity with Der f 7 (the homologous allergen from Dermatophagoides farinae), but its reactivity against Der p 7 is approximately ten‑fold stronger than against Der f 7. The antibody was first described by Shen et al. in Clinical & Experimental Allergy (1995;25(5):416‑22). Clone WH9 is widely used in blocking assays, ELISA, and structural studies aimed at designing hypoallergenic vaccines for house dust mite allergy.
