Adsorption of most proteins can be achieved by mixing equal volume of antigen and adjuvant. Protein left in the supernatant can be measured by protein assay or specific ELISA.
Description
Freund's Adjuvants may be used to produce water-in-oil emulsions of immunogens. The mycobacteria in Complete Freund's adjuvant attract macrophages and other cells to the injection site which enhances the immune response. For this reason, Complete Freund's Adjuvant is used for initial injections and Incomplete Freund's Adjuvant for subsequent boosts. Squalene is a natural 30-carbon organic compound originally obtained for commercial purposes primarily from shark liver oil humans. Squalene as an adjuvant is manufactured by Novartis Classical Adjuvant Combo Pak-1 (contains 10 ml each CFA (#DAGF-051), IFA (#DAGF-053), and Squalene (#DAGF-055); Incomplete Freund's adjuvant/IFA (AV-3015-10), and Squalene (#AV-3020-10) to test adjuvantcy with a given antigen.
