Product Overview
IL-4R alpha protein, Canine (HEK293, His) is the recombinant canine-derived IL-4R alpha protein, expressed by HEK293, with C-6*His labeled tag.
Molecular Weight
Approximately 40-60 kDa
Bio-activity
Measured by its ability to inhibit IL-4-dependent proliferation of TF 1 human erythroleukemic cells.
The ED50 for this effect is ≤18.24 ng/mL in the presence 0.2 ng/mL of Recombinant Human IL-4, corresponding to a specific activity is ≥5.482×104 units/mg.
Endotoxin
<1 EU/μg, determined by LAL method.
Alternative Names
Interleukin-4 receptor subunit alpha; IL-4 receptor subunit alpha; IL-4R subunit alpha; IL-4R-alpha; IL-4RA; CD124
Purity
>95% as determined by reducing SDS-PAGE.
Buffer
Lyophilized from a 0.22 μm filtered solution of PBS, pH 7.4.
Storage
Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
Reconstitution
It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH2O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).
Introduction
Interleukin-4 receptor alpha (IL-4Rα) is a type I transmembrane glycoprotein that serves as the binding subunit for both interleukin-4 (IL-4) and interleukin-13 (IL-13). It plays a crucial role in regulating immune responses, particularly in promoting Th2 cell differentiation, IgE class switching, and alternative macrophage activation. IL-4Rα can form two types of receptor complexes: the type I IL-4 receptor (IL-4Rα with the common γ chain, γc) and the type II receptor (IL-4Rα with IL-13Rα1), mediating IL-4 and IL-13 signaling respectively. Through activation of the JAK-STAT pathway, especially STAT6, IL-4Rα contributes to allergic inflammation, asthma pathogenesis, and various immune-modulatory processes.
