CABT-L6309 exhibits strong reactivity with immobilized β2GPI in ELISA, with a detection limit as low as 20 ng/ml, but only modest interaction with soluble β2GPI in the fluid phase. This characteristic is not a drawback for standard indirect ELISA (where antigen is pre-coated) and may even reduce interference from free antigen; however, assay formats using solution-phase capture may require optimization.
CABT-L6309 specifically recognizes an epitope in Domain I (DI) of human β2GPI, which is the primary target of pathogenic autoantibodies in Antiphospholipid Syndrome (APS). Moreover, when β2GPI is bound to cardiolipin, CABT-L6309 binds not only to human and mouse β2GPI but also to β2GPI from rat, pig, goat, and fetal calf, with varying degrees of reactivity.
