Aspergillus fumigatus is a ubiquitous mold found in soil, decomposing organic matter, and even indoor air. While most healthy individuals can inhale its spores without any ill effects, it can cause a range of serious health problems in immunocompromised patients and those with pre-existing lung conditions. These conditions are collectively known as Aspergillosis. The Anti Aspergillus fumigatus IgG ELISA Quantification Kit is a kit used for quantitative detection of anti Aspergillus fumigatus IgG antibodies in human serum or plasma. By detecting the level of this antibody, it can provide important reference for the auxiliary diagnosis, disease monitoring, and efficacy evaluation of diseases related to Aspergillus fumigatus infection.
Aspergillosis is a group of diseases caused by A. fumigatus and other Aspergillus species. The severity and type of disease depend largely on the patient's immune status and health. The common forms include:
Figure 1. Aspergillosis.
Allergic Bronchopulmonary Aspergillosis (ABPA): An allergic reaction to the fungus, primarily affecting individuals with asthma or cystic fibrosis.
Chronic Pulmonary Aspergillosis (CPA): A long-term, destructive infection of the lungs, often seen in patients with pre-existing lung diseases like tuberculosis or chronic obstructive pulmonary disease (COPD).
Invasive Aspergillosis (IA): A severe and often fatal infection where the fungus spreads from the lungs to other organs. This is a life-threatening condition for severely immunocompromised patients, such as those undergoing chemotherapy, organ transplantation, or with advanced AIDS.
Early and accurate diagnosis of Aspergillosis is crucial for effective treatment and improved patient outcomes.
This kit adopts the principle of enzyme-linked immunosorbent assay (ELISA). The microplate in the reagent kit is pre coated with Aspergillus fumigatus specific antigen. When the serum or plasma sample to be tested is added, the anti Aspergillus fumigatus IgG antibody in the sample can specifically bind to the antigen on the microplate. After washing to remove unbound components, enzyme labeled anti human IgG antibodies are added, which can specifically bind to anti Aspergillus niger IgG antibodies already bound to the antigen. After washing again, add the chromogenic substrate, and the enzyme will catalyze the substrate to undergo a chromogenic reaction. The intensity of the chromogenic reaction is proportional to the concentration of anti Aspergillus niger IgG antibodies in the sample. By measuring the absorbance value of the colorimetric reaction using an enzyme-linked immunosorbent assay (ELISA) reader and comparing it with a standard sample, the content of anti Aspergillus niger IgG antibodies in the sample can be calculated.

| General Description | The fungus Aspergillus belongs to the group of ascomycetes. These organisms develop branched mycelia. They spread through conidiospores which are released from mycelia as extremely resistant lasting spores. 20 species of Aspergillus are human pathogens being opportunistic infectious agents. The most important species is Aspergillus fumigatus. Aspergillus fumigatus can cause multiple allergic and invasive diseases. The pathogenesis of an invasive Aspergillus infection takes place in several steps: The fungal spores are transmitted through the air as a so called "bioaerosol" and are absorbed by the lungs. Due to their size of about 3 μm they reach the lung alveoli. In case of an invasive aspergillosis fungus hyphae penetrate the bronchial mucosa and the surrounding lung parenchyma with the aid of released proteinases. The Aspergillus spores tend to penetrate the blood vessels and settle down in remote tissues after a hematogen dissemination. |
For the diagnosis of chronic pulmonary aspergillosis (CPA) and allergic bronchopulmonary aspergillosis (ABPA), detecting specific anti Aspergillus fumigatus IgG antibodies in the patient's serum is a key serological basis. Elevated (or positive) antibody levels strongly indicate a sustained immune response to Aspergillus fumigatus, which is crucial for distinguishing between colonization and active infections, as well as evaluating treatment efficacy.

Fast Results
ELISA kits offer a much faster turnaround time compared to traditional methods, which can take days. A rapid ELISA can provide results in under two hours, or even as quickly as 30 minutes for some formats.

Direct Sample Testing
Many rapid ELISA kits can be used directly on stool samples, eliminating the need for a separate, time-consuming bacterial culture step. This "culture-independent" method is a major advantage.

Quantitative Results
Unlike qualitative tests that only provide a positive or negative result, this kit provides a specific concentration of IgG antibodies, which can be useful for monitoring disease activity and treatment response.
The Anti-Aspergillus fumigatus IgG ELISA Quantification Kit is an essential tool for clinical laboratories and healthcare providers, aiding in the diagnosis and management of Aspergillosis. By providing a reliable and quantitative measure of the body's immune response to the fungus, it helps clinicians make informed decisions and improve patient care.
The reagent kit should be stored according to the instructions, usually at 2-8 ℃ in the dark to avoid freezing and high temperatures. The shelf life of different components may vary slightly. Generally, unopened reagent kits have a shelf life of about 12 months under specified storage conditions, as indicated on the packaging of the kit. The opened reagents should be used as soon as possible and follow the requirements for storage after opening in the instructions.
At present, this kit is mainly suitable for detecting human serum or plasma samples. Other sample types (such as whole blood, cerebrospinal fluid, etc.) that have not been validated may affect the accuracy of the test results and are not recommended for use.
According to standard operating procedures, the entire testing process from sample preparation to final reading usually takes about 3-4 hours, and the specific time may vary slightly depending on the level of proficiency and laboratory conditions.
