This product (Catalog No. CABT-BL3999) is a rabbit-derived polyclonal antibody raised against human ZSWIM3. This antibody exhibits cross-reactivity with ZSWIM3 from mouse. It has been affinity purified by Protein A. ZSWIM3, also known as zinc finger SWIM-type containing 3, is a protein-coding gene in humans that encodes the zinc finger SWIM domain-containing protein 3, a 696-amino-acid polypeptide featuring a conserved SWIM-type zinc finger domain (residues 531–570) predicted to bind zinc ions and mediate DNA binding as well as protein-protein interactions. Located on the long arm of chromosome 20 at cytogenetic band q13.12 (genomic coordinates GRCh38: 20:45,857,614–45,879,122), the ZSWIM3 gene spans approximately 21.5 kb across three exons and produces a single validated protein isoform (NP_542790.2). The protein is ubiquitously expressed across human tissues, with highest levels observed in the testis (RPKM 2.2) and lymph node (RPKM 1.8), and it has been detected in various fetal tissues during development. ZSWIM3 is part of the conserved ZSWIM gene family across vertebrates, with spatiotemporal expression patterns analyzed in mice, Xenopus, zebrafish, and humans. Functionally, ZSWIM3 plays a critical role in regulating inflammatory responses, particularly in macrophages, where it modulates the TRAF2-mediated NF-κB signaling pathway to suppress ethanol- and lipopolysaccharide (LPS)-induced inflammation. In models of alcohol-induced liver injury, ZSWIM3 expression is downregulated in macrophages via DNA methylation at its promoter (regulated by DNMT3b), leading to exacerbated inflammatory damage; conversely, overexpression of ZSWIM3 alleviates hepatic inflammation and injury. Altered ZSWIM3 expression has also been associated with various liver diseases in both humans and mice, highlighting its involvement in macrophage-mediated immune regulation. Recent studies have also implicated ZSWIM3 in breast cancer progression through lipid metabolism reprogramming and in atherosclerosis hub gene networks.

CABT-BL3999 is supplied as a liquid formulation in Phosphate buffered saline, pH 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
