Intended Use
Human Pancreatic Amylase is a secreted enzyme that is present in saliva and pancreatic secretions in the form of alpha-amylase with 496 amino acids and 56 kDa. Salivary alpha-amylase catalyses the hydrolysis of 1,4-alpha-glycosidic bonds of starch into disaccharide maltose, trisaccharide maltotriose, and small dextrins. Pancreatic alpha-amylase continues the hydrolysis of starch into disaccharides and trisaccharides which are converted by alpha-glucosidases to absorbable glucose, fructose, and galactose in the small intestine. The serum amylase concentration is increased in acute pancreatitis, and ovarian tumors. By retardation of carbohydrate digestion, the amylase inhibitor has anti-obesity and anti-diabetes effects and can control postprandial hyperglycemia in type 2 diabetes. Salivary alpha-amylase has been proposed as a stress biomarker in autonomic/sympathetic nervous system.

1. Human Amylase Microplate: A 96-well polystyrene microplate (12 strips of 8 wells) coated with a polyclonal antibody against human Amylase. 96 (8×12) wells
2. Sealing Tapes: Pressure-sensitive sealing tapes that can be cut to fit the format of the individual assay. 3 slides
3. Human Amylase Standard (Lyophilized): Human Amylase in a buffered protein base. 160 mU
4. Biotinylated Human Amylase Antibody (100×): A 100-fold concentrated biotinylated polyclonal antibody against Amylase. 80 μL
5. MIX Diluent Concentrate (10×): A 10-fold concentrated buffered protein base. 30 mL
6. Wash Buffer Concentrate (20×): A 20-fold concentrated buffered surfactant. 30 mL × 2
7. Streptavidin-Peroxidase Conjugate (SP Conjugate): A 100-fold concentrate. 80 μL
8. Chromogen Substrate: A ready-to-use stabilized peroxidase chromogen substrate tetramethylbenzidine. 8 mL
9. Stop Solution: A 0.5 N hydrochloric acid to stop the chromogen substrate reaction. 12 mL
Storage
1. Upon arrival, immediately store components of the kit at recommended temperatures up to the expiration date.
2. Store SP Conjugate and Biotinylated Antibody at -20°C.
3. Store Microplate, Diluent Concentrate (10×), Wash Buffer, Stop Solution, and Chromogen Substrate at 2-8°C.
4. Unused microplate wells may be returned to the foil pouch with the desiccant packs and resealed. May be stored for up to 30 days in a vacuum desiccator.
5. Diluent (1×) may be stored for up to 30 days at 2-8°C.
6. Store Standard at 2-8°C before reconstituting with Diluent and at -20°C after reconstituting with Diluent.
Precision
Intra-assay precision was determined by testing replicates of three plasma samples in one assay.
Inter-assay precision was determined by testing three plasma samples in twenty assays.
Detection Range
0.313 to 20.00 mU/mL
Detection Limit
The minimum detectable dose of Human Amylase as calculated by 2SD from the mean of a zero standard was established to be 0.15 mU/mL.
Standard Curve
The typical data is provided for reference only. Individual laboratory means may vary from the values listed. Variations between laboratories may be caused by technique differences.

The curve is used for illustration only. A standard curve should be generated each time the assay is performed.
