Intended Use
The Human E-Cadherin / CDH1 / E-cad / CD324 ELISA Pair Set is for the quantitative determination of Human E-Cadherin / CDH1 / E-cad / CD324.
This ELISA Pair Set contains the basic components required for the development of sandwich ELISAs.
Contents of Kit
Capture Antibody: 1 mg/mL of mouse anti-Human E-Cadherin / CDH1 / E-cad / CD324 monoclonal antibody (in PBS, pH 7.4). Dilute to a working concentration of 2 μg/mL in PBS before coating.
Detection Antibody: 0.2 mg/mL of rabbit anti-Human E-Cadherin / CDH1 / E-cad / CD324 polyclonal antibody conjugated to horseradish-peroxidase (HRP) (in PBS, 50 % HRP-Protector, pH 7.4, store at 4°C). Dilute to working concentration of 0.25 μg/mL in Dilution Buffer before use.
Standard: Each vial contains 115 ng of recombinant Human E-Cadherin / CDH1 / E-cad / CD324. Reconstitute with 1 mL Dilution Buffer. After reconstitution, store at -20°C to -80°C in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in Dilution Buffer, and a high standard of 1600 pg/mL is recommended.
Storage
Capture Antibody: Aliquot and store at -20°C to -80°C for up to 6 months from date of receipt. Avoid repeated freeze-thaw cycles.
Detection Antibody: Store at 4°C and protect it from prolonged exposure to light for up to 6 months from date of receipt. Do not freeze!
Standard: Store lyophilized standard at -20°C to -80°C for up to 6 months from date of receipt. Aliquot and store the reconstituted standard at -80°C for up to 1 month. Avoid repeated freezethaw cycles.
Sensitivity
The minimum detectable dose of Human COMP / TSP5 was determined to be approximately 39.06 pg/ml.
This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.
General Description
Cadherins are calcium-dependent cell adhesion proteins which preferentially interact with themselves in a homophilic manner in connecting cells, and thus may contribute to the sorting of heterogeneous cell type. E-cadherin (E-Cad), also known as CDH1 and CD324, is a calcium-dependent cell adhesion molecule the intact function of which is crucial for the establishment and maintenance of epithelial tissue polarity and structural integrity. Mutations in CDH1 occur in diffuse type gastric cancer, lobular breast cancer, and endometrial cancer. In human cancers, partial or complete loss of E-cadherin expression correlates with malignancy. During apoptosis or with calcium influx, E-Cad is cleaved by the metalloproteinase to produce fragments of about 38 kDa (E-CAD/CTF1), 33 kDa (E-CAD/CTF2) and 29 kDa (E-CAD/CTF3), respectively. E-Cad has been identified as a potent invasive suppressor, as downregulation of E-cadherin expression is involved in dysfunction of the cell-cell adhesion system, and often correlates with strong invasive potential and poor prognosis of human carcinomas.
