This product (Catalog No. DPABH-18342) is a rabbit-derived polyclonal antibody raised against human ZDHHC14. This antibody exhibits cross-reactivity with ZDHHC14 from mouse and rat. It has been affinity purified by Protein A. ZDHHC14 is a protein-coding gene located on the long arm of human chromosome 6 at position 6q25.3 that encodes zDHHC palmitoyltransferase 14, an enzyme belonging to the DHHC family of palmitoyl acyltransferases (PATs) responsible for catalyzing the S-palmitoylation of substrate proteins, a reversible post-translational modification that influences protein trafficking, stability, and function. This transferase is characterized by a conserved structure featuring multiple transmembrane domains, a catalytic DHHC cysteine-rich domain, and a C-terminal LSSV motif that enables interaction with PDZ domain-containing scaffold proteins such as PSD-93 (DLG2), facilitating targeted palmitoylation in cellular compartments like the endoplasmic reticulum and Golgi apparatus. In neuronal cells, ZDHHC14 plays a pivotal role in regulating excitability by palmitoylating and promoting the clustering of Kv1-family voltage-gated potassium channel subunits (e.g., KCNA1, KCNA2, and KCNA4) and associated scaffolds at the axon initial segment (AIS), which is essential for proper action potential modulation; disruption of this process, as seen in knockout models, leads to reduced potassium currents and increased neuronal firing. Expression of ZDHHC14 is ubiquitous across human tissues, with elevated levels in the brain (particularly the hippocampus) and endometrium, and it is active during fetal development in organs such as the adrenal gland, heart, and kidney. Although no monogenic human disorders are directly attributed to ZDHHC14 variants, the gene resides within a chromosomal region (6q24-q25) frequently deleted in conditions like chromosome 6q24-q25 deletion syndrome, and its downregulation—without accompanying mutations or promoter hypermethylation—occurs in testicular germ cell tumors and prostate cancer, where it exhibits tumor-suppressive effects by inhibiting cell viability, inducing apoptosis, and suppressing xenograft tumor growth in mouse models.

DPABH-18342 is supplied as a liquid formulation in Phosphate buffered saline, pH 7.4, 150mM NaCl, 0.02% sodium azide and 50% glycerol. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
