This product (Catalog No. DPABH-22464) is a rabbit-derived polyclonal antibody raised against human SH2D3A. It has been affinity purified by Protein A. SH2D3A is a human gene located on chromosome 19p13.3 that encodes an adaptor protein known as SH2 domain-containing protein 3A, or novel SH2-containing protein 1 (NSP1), which facilitates signal transduction by linking integrin and receptor tyrosine kinase signaling to the c-Jun N-terminal kinase (JNK)/stress-activated protein kinase (SAPK) pathway. The protein, consisting of 576 amino acids with a molecular weight of approximately 64 kDa, features an SH2 domain for phosphotyrosine binding and a Ras guanine nucleotide exchange factor (RasGEF) domain that activates Ras-like small GTPases, contributing to processes such as cytoskeleton dynamics and intracellular trafficking. Expressed broadly but notably in tissues like the placenta, pancreas, kidney, lung, stomach, and colon, SH2D3A interacts with proteins such as p130-Cas (BCAR1) and is responsive to stimuli including insulin, epidermal growth factor (EGF), and fibronectin, leading to its tyrosine phosphorylation and selective activation of JNK1 without affecting ERK2. While primarily studied for its role in JNK cascade regulation, which influences cellular responses like proliferation and apoptosis, emerging research suggests potential involvement in cancer progression, such as hypoxia-induced self-replication in lung cancer stem cells, though no specific disease associations have been firmly established. The gene produces multiple isoforms, with at least 15 variants identified, varying in their SH2 and RasGEF domains, and belongs to a family of adaptors (including BCAR3 and SHEP1) implicated in cell adhesion, migration, and morphology.

DPABH-22464 is supplied as a liquid formulation in pH7.4 PBS, 0.05% sodium azide, 40% Glycerol. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
