This product (Catalog No. DPABH-04025) is a rabbit-derived polyclonal antibody raised against human GCLM, targeting the amino acid region 40–251 of the protein. This antibody exhibits cross-reactivity with Human GCLM from mouse and rat. It has been affinity purified by Protein A. Glutamate-cysteine ligase modifier subunit (GCLM) is a protein-coding gene that encodes the regulatory subunit of glutamate-cysteine ligase, the rate-limiting enzyme in the biosynthesis of glutathione, a critical cellular antioxidant. Located on the short arm of human chromosome 1 at position 1p22.1, GCLM forms a heterodimer with the catalytic subunit (GCLC) to enhance enzymatic activity and modulate glutathione production in response to oxidative stress. This subunit plays an essential role in cellular redox homeostasis, detoxification, and protection against oxidative damage, with deficiencies linked to conditions such as hemolytic anemia and increased susceptibility to environmental toxins. Variations in GCLM expression have been associated with metabolic disorders, including myocardial infarction and abdominal obesity-metabolic syndrome, highlighting its impact on cardiovascular and inflammatory pathways. Recent research has also implicated elevated GCLM levels in colorectal cancer chemoresistance, where its nuclear translocation promotes tumor survival under therapeutic stress.
DPABH-04025 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-04025 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
