This product (Catalog No. DPABH-29039) is a rabbit-derived polyclonal antibody raised against human CAPN2, targeting the amino acid region 45–514 of the protein. This antibody exhibits cross-reactivity with Human CAPN2 from mouse and rat. It has been affinity purified by Protein A. Calpain-2, also known as m-calpain or CAPN2, is a ubiquitous intracellular cysteine protease that belongs to the calpain family of calcium-dependent enzymes, characterized by its role in limited, non-degradative proteolysis to regulate protein function rather than complete breakdown. It exists as a heterodimer consisting of an 80 kDa catalytic subunit (encoded by the CAPN2 gene) containing the active site and EF-hand calcium-binding domains, and a 28-30 kDa regulatory subunit (CAPNS1, shared with calpain-1) that provides stability and facilitates heterodimerization. Activation requires millimolar concentrations of Ca²⁺, which induce a conformational change to expose the catalytic triad (cysteine, histidine, asparagine), distinguishing it from the related calpain-1 (μ-calpain), which activates at micromolar Ca²⁺ levels.
DPABH-29039 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:200), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-29039 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
