General Notes
This product (Catalog No. DPABH-16905) is a rabbit-derived polyclonal antibody raised against human C9, targeting the amino acid region 22–265 of the protein. It has been affinity purified by Protein A. Complement component 9 (C9) is a 71-kDa glycoprotein and the terminal protein in the complement cascade of the innate immune system, essential for completing the membrane attack complex (MAC, also known as C5b-9) that forms lytic pores in pathogen membranes to facilitate cell destruction. Primarily synthesized in the liver and circulating in plasma at approximately 60 μg/mL, C9 binds to the C5b-8 complex on target cell surfaces, where multiple C9 monomers (typically 12–18) polymerize into a ring-like structure with a central pore diameter of about 10 nm, penetrating the lipid bilayer to cause osmotic lysis, particularly effective against Gram-negative bacteria like Neisseria meningitidis. As a member of the membrane attack complex/perforin (MACPF) superfamily, C9 undergoes conformational changes during activation, with its amphiphilic structure—featuring a hydrophilic N-terminal MACPF domain and hydrophobic C-terminal regions—enabling deep insertion into hydrocarbon cores of membranes, which is critical for the cytolytic activity that defends against infections while regulating inflammation through immune complex clearance and cytokine release.
DPABH-16905 is suitable for use in ELISA (1:4,000–1:8,000), IHC (1:50–1:100), Western blot (WB) (1:1,000–1:4,000) applications.
DPABH-16905 is supplied as a liquid formulation in 0.01M PBS, pH 7.4, 50% Glycerol, 0.05% Proclin 300. For long-term storage, keep at −20 °C. For short-term use, store at 2–8 °C. Avoid repeated freeze–thaw cycles to maintain antibody integrity and performance.
